When you replay a mission, you can also repeat the mission briefing but you won't be able to watch any cutscene or conversation that may have occurred during your first playthrough of that mission. Simply put, the answer is both yes and no. While on a mission you simply can't customize or change your build. However, if you die while on a mission, you can change your build before respawning.
